Drug Class
Aminopenicillin with beta-lactamase inhibitor
Indications
- Respiratory tract infections (sinusitis, pneumonia, bronchitis)
- Otitis media
- Urinary tract infections
- Skin and soft tissue infections
- Bone and joint infections
Dosage
- Adults: 500/125 mg every 8 hours OR 875/125 mg every 12 hours
- Children: 20–45 mg/kg/day of amoxicillin component, divided every 8–12 hours
Note: Adjust dose in renal impairment and according to infection severity.
Route of Administration
Oral (tablet, suspension), IV for severe infections
Mechanism of Action
Amoxicillin inhibits bacterial cell wall synthesis, while clavulanic acid inhibits beta-lactamase enzymes, extending amoxicillin's spectrum against resistant bacteria.
Common Adverse Effects
- Diarrhea
- Nausea, vomiting
- Skin rash
- Abdominal discomfort
Serious Adverse Effects
- Anaphylaxis
- Severe hypersensitivity reactions
- Clostridioides difficile–associated diarrhea
- Hepatic dysfunction (rare)
Contraindications
- Hypersensitivity to penicillins or beta-lactamase inhibitors
- History of cholestatic jaundice or hepatic dysfunction with this drug
Precautions
- History of allergy or asthma
- Renal impairment (dose adjustment needed)
- Monitor for diarrhea and liver function if prolonged use
Drug Interactions
- Allopurinol – increased risk of rash
- Oral contraceptives – effectiveness may be reduced
- Warfarin – may increase anticoagulant effect
Pregnancy & Lactation
Generally safe in pregnancy and breastfeeding if used as directed.
Patient Counseling
- Complete the full prescribed course
- Take with food to reduce gastrointestinal upset
- Report rash, diarrhea, or jaundice immediately
- Do not share medication with others
